Effect of drug-coformer interactions on drug dissolution from a coamorphous in mesoporous silica

In this study, the molecular state of ritonavir (RTN)-saccharin (SAC) coamorphous incorporated into mesoporous silica by solvent evaporation and effect SAC on RTN dissolution from mesopores were investigated. The amorphization RTN-SAC was confirmed as a halo pattern in powder X-ray diffraction measurements single glass transition event modulated differential scanning calorimetry (MDSC) curve. 13C solid-state NMR spectroscopy revealed hydrogen bond between thiazole nitrogen amine proton SAC. not found MDSC curve, indicating that monomolecularly mesopores. Solid-state suggested co-incorporation decreased local mobility group via formation. 1:1 retained halo-patterns after 30 d storage, even under high temperature humidity conditions. test, maintained supersaturation for longer time than amorphous silica. This study demonstrated drug-coformer interaction within can significantly improve drug dissolution.
